Mutation location is the key to prognosis
Feb. 28, 2013 ? The three most important factors in real estate are location, location, location, and the same might be said for mutations in the gene MECP2, said researchers at Baylor College of Medicine and the Jan and Dan Duncan Neurological Research Institute at Texas Children's Hospital in a report in the journal Cell.
"Where a mutation occurs can affect the severity of the symptoms of the disease," said Dr. Huda Zoghbi, professor of molecular and human genetics at BCM and director of the NRI. Zoghbi, corresponding author of the report, found the MECP2 gene in 1999 and confirmed that deficiency in the protein causes Rett syndrome, a post natal genetic disease that mainly affects girls.
Symptoms influenced by gene location
In the study, she and her colleagues relied on data from rare male patients with disruptions in MECP2 that showed that severity of symptoms could be influenced by the location of the gene mutation. The few boys with this disorder fell into two broad categories: Those who suffered severe brain disease and death before age 4 and those who lived for decades with symptoms similar to that of Rett or developmental delay and other disorders similar to those seen in autism.
Looking at the placement of the mutations in the boys, they hypothesized there was a distinct difference in symptoms seen in boys who had mutations at amino acid 270 in the protein and those who had mutations only slightly farther along, at amino acid 273. The protein is truncated or shorter in those with amino acid 270 mutations than those with the mutation at amino acid 273.
After Steven Baker, a graduate student in the Program in Developmental Biology at BCM, generated and characterized mice that had mutations at the two sites of the protein, he found that mice who had mutations at amino acid 273 lived longer and developed symptoms later than those mice who had mutations at amino acid 270 or those who lacked the MeCP2 protein all together (knock-out mice).
Disruption of topological feature
One reason for the differences could be that the mutation at amino acid 270 disrupts a key topological feature of the DNA -- an AT-Hook domain that is a DNA binding motif. By disrupting this domain, the mutation could affect the way the protein bind the DNA and make the already truncated protein much less effective.
"The participation of patients and their families with Rett researchers really helped us to key in on regions of MeCP2 that are critical for its function," said Baker, who is also an M.D./Ph.D. student in BCM's Medical Student Training Program.
The researchers propose a model for this DNA binding in which MeCP2 binds to sites across the genome. In some spots where this occurs, the protein manipulates the structure of the nearby chromatin.
Chromatin architectural factor
"We think that one function of MeCP2 (the protein associated with the gene) is to alter the architecture of chromatin (the mass of proteins and DNA found in the nucleus of the cell)," said Zoghbi, who is also Howard Hughes Medical Institute investigator.
Baker said, "The picture of MeCP2 as a chromatin architectural factor is emerging from the combined efforts of many laboratories. Understanding how MeCP2 modifies chromatin structure will ultimately allow us to understand why it is so important for neuronal health."
Others who took part in this work include Lin Chen, Angela Dawn Wilkins, Peng Yu and Olivier Lichtarge, all of BCM.

Texas man who set ex-girlfriend on fire set to die
HUNTSVILLE, Texas (AP) - A federal appeals court has refused to halt the scheduled Thursday evening execution of a Texas man convicted of killing his ex-girlfriend by tossing a cup of gasoline on her and then setting her on fire.
The New Orleans-based 5th U.S. Circuit Court of Appeals, in a brief ruling Wednesday evening, rejected an appeal filed by an attorney for 48-year-old Carl Blue.
Blue faces lethal injection in Huntsville for the 1994 attack that killed 38-year-old Carmen Richards-Sanders at her apartment in Bryan.
Blue's lawyer, Michael Charlton, says he's taking Blue's appeal to the U.S. Supreme Court.

French, Malian forces fight Islamist rebels in Gao
GAO, Mali (Reuters) - French and Malian troops fought Islamists on the streets of Gao and a car bomb exploded in Kidal on Thursday, as fighting showed little sign of abating weeks before France plans to start withdrawing some forces.
Reuters reporters in Gao in the country's desert north said French and Malian forces fired at the mayor's office with heavy machineguns after Islamists were reported to have infiltrated the Niger River town during a night of explosions and gunfire.
French Defense Minister Jean-Yves Le Drian told a news conference in Brussels that Gao was back under control after clashes earlier in the day.
"Malian troops supported by French soldiers killed five jihadists and the situation is back to normal," he said.
In Kidal, a remote far north town where the French are hunting Islamists, residents said a car bomb killed two. A French defense ministry source reported no French casualties.
French troops dispatched to root out rebels with links to al Qaeda swiftly retook northern towns last month. But they now risk being bogged down in a guerrilla conflict as they try to help Mali's weak army counter bombings and raids.
"There was an infiltration by Islamists overnight and there is shooting all over the place," Sadou Harouna Diallo, Gao's mayor, told Reuters by telephone earlier in the day, saying he was not in his office at the time.
Gao is a French hub for operations in the Kidal region, about 300 km (190 miles) northeast, where many Islamist leaders are thought to have retreated and foreign hostages may be held.
"They are black and two were disguised as women," a Malian soldier in Gao who gave his name only as Sergeant Assak told Reuters during a pause in heavy gunfire around Independence Square.
Six Malian military pickups were deployed in the square and opened fire on the mayor's office with the heavy machineguns. Two injured soldiers were taken away in an ambulance.
French troops in armored vehicles later joined the battle as it spilled out into the warren of sandy streets, where, two weeks ago, they also fought for hours against Islamists who had infiltrated the town via the nearby river.
Helicopters clattered over the mayor's office, while a nearby local government office and petrol station was on fire.
A Gao resident said he heard an explosion and then saw a Malian military vehicle on fire in a nearby street.
Paris has said it plans to start withdrawing some of its 4,000 troops from Mali next month. But rebels have fought back against Mali's weak and divided army, and African forces due to take over the French role are not yet in place.
Islamists abandoned the main towns they held but French and Malian forces have said there are pockets of Islamist resistance across the north, which is about the size of France.
CAR BOMB
Residents reported a bomb in the east of Kidal on Thursday.
"It was a car bomb that exploded in a garage," said one resident who went to the scene but asked not to be named.
"The driver and another man were killed. Two other people were injured," he added.
A French defense ministry official confirmed there had been a car bomb but said it did not appear that French troops, based at the town's airport, had been targeted.
Earlier this week, a French soldier was killed in heavy fighting north of Kidal, where French and Chadian troops are hunting Islamists in the Adrar des Ifoghas mountains, which border Algeria.
Operations there are further complicated by the presence of separatist Tuareg rebels, whose rebellion triggered the fighting in northern Mali last year but were sidelined by the better-armed Islamists.
Having dispatched its forces to prevent an Islamist advance south in January, Paris is eager not to become bogged down in a long-term conflict in Mali. But their Malian and African allies have urged French troops not to pull out too soon.

Twin bombings kill 14, wound more in south India
HYDERABAD, India (AP) ? A day after two bicycle bombs killed 14 people and wounded more than 100, investigators into India's worst bombing in more than a year searched Friday for possible links to anger over the execution of a Muslim militant.
The bombs exploded minutes apart late Thursday in a crowded shopping area in the southern city of Hyderabad ? one of them near a cinema and one near a bus station. The blasts shattered storefronts, scattered food and plates from roadside restaurants and left tangles of dead bodies. Passersby rushed the wounded to hospitals.
"This is a dastardly attack; the guilty will not go unpunished," Prime Minister Manmohan Singh said. He appealed to the public to remain calm.
India has been in a state of alert since Mohammed Afzal Guru, a Kashmiri, was hanged in a New Delhi jail nearly two weeks ago after being convicted of involvement in a 2001 attack on India's Parliament. The assault killed 14 people, including five of the gunmen.
Many in Indian-ruled Kashmir believe Guru did not receive a fair trial, and the secrecy with which the execution was carried out fueled anger in a region where anti-India sentiment runs deep.
Home Minister Sushilkumar Shinde said there was a general alert about the possibility of an attack somewhere in India for the past three days. "But there was no specific intelligence about a particular place," he said.
The bombs were attached to two bicycles about 150 meters (500 feet) apart in Hyderabad's Dilsukh Nagar district, Shinde told reporters in New Delhi. He said 14 people died and 119 others were injured, six of them critically.
Top state police officer V. Dinesh Reddy said improvised explosive devices with nitrogen compound were used in the blasts.
Mahesh Kumar, a 21-year-old student, was heading home from a tutoring class when a bomb went off.
"I heard a huge sound and something hit me, I fell down, and somebody brought me to the hospital," said Kumar, who suffered shrapnel wounds.
Hyderabad, a city of 10 million in the state of Andhra Pradesh, is a hub of India's information technology industry and has a mixed population of Muslims and Hindus.
"This (attack) is to disturb the peaceful living of all communities in Andhra Pradesh," said Kiran Kumar Reddy, the state's chief minister.
The explosions were the first major bomb attack to hit India since a September 2011 blast outside the High Court in New Delhi killed 13 people. The government has been heavily criticized for its failure to arrest the masterminds behind previous bombings.
Officials from the National Investigation Agency and commandos of the National Security Guards arrived from New Delhi to help with the investigation in Hyderabad.
The United States, whose Secretary of State John Kerry was meeting Thursday in Washington with Indian Foreign Secretary Ranjan Mathai, condemned the attack.
"The United States stands with India in combating the scourge of terrorism and we also prepared to offer any and all assistance Indian authorities may need," State Department spokeswoman Victoria Nuland told a news briefing.
Rana Banerji, a former security official, said India remains vulnerable to such attacks because there is poor coordination between the national government and the states. Police reforms are also moving very slowly and the quality of intelligence gathering is poor, he said.
"The concept of homeland security should be made effective, on a war footing," he said.
Rajnath Singh, the president of main opposition Bharatiya Janata Party, demanded a thorough probe into the blasts. His party called for a general strike in the state on Friday.

Climate change effect on plant communities is buffered by large herbivores
Can existing ecological communities persist intact as temperatures rise? This is a question of increasing relevance in the field of climate change and is the focus of a new study to be published in the journal Proceedings of the Royal Society of London on 20 February. The study suggests that the answer to this question may have as much to do with the biological interactions that shape communities as with the effects of climate change itself.
The study's insights are based on a novel approach by Eric Post, a Penn State University professor of biology, who simulated climate change and integrated the effects of large, plant-eating mammals in a 10-year arctic field experiment. The results of the research suggest that plant communities in the Arctic are more likely to resist destabilization by climate change if populations of caribou, musk ox, and other large herbivores remain intact. "The study demonstrates that grazing by these large herbivores maintains plant species diversity, while warming reduces it," Post said. "Plant communities with lower diversity display a greater tendency toward instability under warming, a pre-cursor to the loss of such communities."
Post explained that climate-change research in the 1980s and 1990s was focused primarily on how fluctuations in such factors as temperature, precipitation, and nutrient availability directly affected plant communities. "That research was important and enlightening, but what it did not emphasize were the indirect effects of climate change -- how interactions among species may shape the responses of those species to warmer temperatures," Post said. "If the planet continues to warm by 1.5-to-3.0 degrees Celsius over the next century, as the models predict, we need to know not only what the warming will do to plants and animals directly, but also how species' interactions may influence those effects of warming."
Post began the study in a remote, low-Arctic plant community near Kangerlussuaq, Greenland in 2002. To simulate the effects of the 1.5-to-3.0-degrees-Celsius warming that is predicted to occur over the next century, he erected special warming chambers -- cone-shaped hollow structures that create a greenhouse effect. Some areas on which these warming chambers were placed were left open to grazing by caribou and musk ox -- two ecologically important large herbivores in the Arctic -- while separate 800-square-meter areas that also received warming chambers were fenced off to exclude the animals. In this way, Post created two very different environments: one in which plants and herbivores continued to live together as the temperatures climbed within the warming chambers; the other in which the animals were not present and the plants were left ungrazed.
"The study tested a classic ecological hypothesis, but with a new angle," Post said. "Ecologists have argued for decades over whether species-rich plant communities are more stable, and, hence, persistent in the face of environmental disturbance, than species-poor communities. This study added a layer of complexity by asking whether large herbivores contribute to the diversity-stability relationship in a climate-change context."
After 10 years of careful observation of the Kangerlussuaq, Greenland plant communities, Post found that the grazed and ungrazed sections of land did indeed fare quite differently in their responses to warmer temperatures. "This study confirmed that caribou and musk ox act as a buffer against the degradative effects of warming on plant species diversity," Post said. He found that shrubs such as willow and birch became the dominant plants in response to warming where the herbivorous animals were excluded from the ecosystem. "When these shrubs expand in the plant community, they tend to shade their neighbors, and the build-up of leaf litter around the shrubs tends to cool the soil surface, reducing the availability of soil nutrients for other plants," Post said. "As a result, shrubs can quickly out-compete other plants and reduce species diversity in the process. On the other hand, in those areas where caribou and musk ox were able to graze freely, shrub responses to warming were muted, and species diversity within the plant community was maintained."
Post said the take-home message from his study is that, in a warming climate, intact populations of large herbivores may be crucial to the maintenance of plant-community diversity and to the persistence of existing plant communities. "What this experiment suggests is that factors that threaten the persistence of large herbivores may threaten the plant communities they exist in, as well. Conservation of these herbivores in the rapidly changing Arctic will require careful mediation of interacting stressors such as human exploitation, mineral extraction, and the direct effects of climate change," Post said.
Post said that the next step in his research will be to study the contribution of plant diversity to long-term stability of carbon dynamics in the atmosphere and in the soil.

Air Canada, Lufthansa, United tie-up likely to win EU approval: source
BRUSSELS ? Lufthansa, United Airlines and Air Canada are set to win approval from the European Union?s competition regulator for their transatlantic tie-up with their offer to give up airport slots, a person familiar with the matter said on Wednesday.
The three airlines last year proposed giving up landing and take-off slots at Frankfurt and New York?s airports following a three-year investigation by the European Commission into their revenue-sharing, pricing and capacity pacts.
The EU antitrust authority sought feedback from rivals and third parties in December last year.
?The Commission is likely to accept the airlines? proposal with some fine tuning,? said the person who declined to be named because of the sensitivity of the matter.
Regulatory approval means the airlines will not be found guilty of anti-trust infringement and will not face fines which could reach 10% of a company?s global revenues.
The Commission spokesman for competition policy could not immediately be reached for comment.
The three carriers are all members of the Star Alliance of airlines, the biggest such marketing group with 27 members accounting for a quarter of global airline capacity.
Such alliances were set up in the 1990s to enable airlines to extend their apparent networks without having to overcome the industry?s peculiar restrictions on international mergers.
This would be the second airline network cleared by the Commission. It approved a tighter transatlantic tie-up between British Airways, American Airlines and Iberia in July 2010.
All three are members of the broader oneworld alliance.
The EU watchdog is also currently investigating a transatlantic joint venture between Air France-KLM, Alitalia and Delta Airlines.

Ding-dong, Hotmail's dead: All accounts switching to Outlook.com by summer

It's official: Outlook.com will fully replace Hotmail as Microsoft's webmail service. The company will begin to auto-update accounts, and hopes they will be fully migrated from Hotmail to Outlook.com by this summer.
The move was spurred by surprise growth in the Outlook.com mail service, which has amassed 60 million active users in just six months. Microsoft will also remove the "customer preview" label on the product, and launch a multimillion-dollar marketing campaign.

I know the first thing you're thinking: "Does this mean my Hotmail address will just go away?" No. It just means that when you go to hotmail.com, you will be re-routed to Outlook.com, and when you log in on the Web, you will get the Outlook.com experience. You can keep your @hotmail.com email address forever, but you can also use that account to create multiple new @outlook.com email addresses, too, if you so desire.
And let me tell you, you shouldn't wait for Microsoft to switch your old Hotmail service over to Outlook.com. I am an unabashed Hotmail hater, but who can forgive the original webmail service for being so far behind the times? Even when Microsoft spent millions on a "new Hotmail" ad campaign a scant two years ago, nobody was fooled: You still had to refresh the thing every time you wanted to know if you had mail.
Enter Outlook.com, which really can give Gmail a run for its money. It's a very smart service with a very streamlined design, tasteful social integration and auto organization features such as inbox "sweep" and scheduled cleanup. Because of this last bit, it's ideal for use either as a main email or as a "spam account," the kind you provide to online retailers and other data collectors.
(Switching your account over takes almost no effort: Just log into Hotmail then click Settings at the top right ? you will see the option to convert to Outlook.)
Though Microsoft was pleased at the sudden growth of Outlook.com users, I grilled David Law, director of product management for Outlook.com, about how many of the 60 million users were just converts from Hotmail's existing 350 million or so accounts. My supposition is that this represents almost all of them, but while Law wouldn't tell me the number, he did say I would be surprised how many were totally fresh.
Law was forthcoming about a different statistic, one that demonstrates Microsoft's target: About one third of the current Outlook.com users are ? or, at least, were ? also Gmail users.
Outlook.com is a very welcome email option, and ? when partnered with the company's SkyDrive cloud storage services ? a sign that Microsoft may well regain some traction with consumers looking for reliable Web services.
One of its only obvious problems is that of branding: Because it's called "Outlook," many people (naturally) assume that it is some kind of Web client for managing any email account. It's not. It's a free service, like Gmail or Yahoo mail or, yes, Hotmail, that provides you with an email account. So don't go asking if you can create an Outlook.com account and then add your corporate email to it, because that's like asking if you can take Google Maps and stick Mapquest into it.

A bill in the legislature would make tanning beds illegal for anyone under the age of 18, even with a parent's permission.
The bill's sponsors claim it aims to protect children from the "harmful effects of exposure to ultraviolet radiation associated with tanning devices."
Monica Kelley does not like the idea for two reasons.
Kelley owns a tanning salon in Olympia called "Tanfastic Electric Beach." She said the law would hurt the business she bought earlier this year. She estimates 20 percent of her customers are under 18.
Kelley also does not think using tanning beds is unhealthy and she thinks parents should be able to decide if their younger teens want to get a tan.

Show jumper Shane Sweetnam has no intention of just making up the numbers when he contests his first World Cup final next April in Sweden.
The US-based Corkman steered Amaretto d?Arco to third place on Saturday night in the $125,000 World Cup qualifier in Florida to copperfasten his place in the final and yesterday said he would be as competitive as ever in looking to give Ireland its first World Cup title.?This is the first year I really tried to qualify for the final. It was a goal for the second half of the year and now we can concentrate on the trip.
?The World Cup is a true championship and I am going there to compete properly. My horse has plenty of experience and we will give it a good go. He was good at the European Championships in 2011 ? he was fourth on the last day of competition ? so he?s shown he can compete at the highest level over a few days.?
Sweetnam also welcomed Robert Splaine?s reappointment last week as Ireland show jumping manager.
?I think he?s done a good job and he ensured we stayed at the top level in the nations cup league over the past few years. I think the success can continue and we can build on that and I?d like to think I can play my part.?
Meanwhile, Daniel Coyle recorded back-to-back wins in the HSI/Blue Grass Spring Tour with victory in the second leg at Ravensdale, Co Louth, yesterday.
Again it was Linzi Johnston?s Uptown Girl that ensured the Derry rider walked away with the top points to give him the perfect start to defending his title.
The only other clear in the five-horse jump-off came from Sophie Dalm and Unique, while Coyle also provided the fastest of the four-faulters with Special Leap.

Facebook to open up 100 jobs in Dublin to expand its European hub
Facebook is bringing 100 new positions to Ireland as part of its ongoing expansion in Europe. Irish Broadcaster RTE says that this will add to Facebook?s team of 400 people in the country.
The new positions are already showing up on Facebook?s recruitment pages. Positions range from corporate communications, developer relations, systems, user operations, sales and many more.
Ireland?s Minister for Jobs, Enterprise and Innovation, Richard Bruton is encouraging the expansion and feels that it helps Dublin stand out as a centre for technology. He said; ?Today?s announcement of a significant expansion by one of the biggest companies on the internet, with the creation of 100 high-end jobs, is a major endorsement of the tech environment in Dublin and further confirmation of our city?s status as the internet capital of Europe.?
Indeed Ireland is seeing some of the most recognisable names online expand their companies in the country. Just last week the auction giant eBay announced the creation of a further 450 jobs in Ireland and last year PayPal said it would be adding 1,000 positions?over a four-year period.
Bruton is pinning hopes to change Ireland?s economic outlook on the ICT sector. Earlier this month at the Future Jobs Forum in Dublin he pointed to the European gap between graduates and technology positions to be filled and said that technology is a changing force across many markets. ?There?s no doubt when you look at it the ICT sector and what it is bringing to the table, it is dramatically changing the face of a lot of sectors, democratising start-ups who don?t have the same barriers they used to, it is compressing the world market,? he said.
Supporting the burgeoning startup culture in Ireland is bound to bring benefits and hope for the economy, but a few big names opening up more positions in the tech sector is bound to be welcome news.

Reduced sea ice disturbs balance of greenhouse gases
Feb. 18, 2013 ? The widespread reduction in Arctic sea ice is causing significant changes to the balance of greenhouse gases in the atmosphere. This is shown in a new study conducted by researchers from Lund University in Sweden, among others.
According to the study, the melting of sea ice in the Arctic has a tangible impact on the balance of greenhouse gases in this region, both in terms of uptake and release. The researchers have studied the greenhouse gases carbon dioxide and methane both in the tundra and in the Arctic Ocean.
"Changes in the balance of greenhouse gases can have major consequences because, globally, plants and the oceans absorb around half of the carbon dioxide that humans release into the air through the use of fossil fuels. If the Arctic component of this buffer changes, so will the amount of greenhouse gases in the atmosphere," says Dr Frans-Jan Parmentier, a researcher at Lund University, Sweden.
He has carried out the research study together with a number of colleagues both from Lund University and from Denmark, Greenland, Canada and the USA. The researchers observed that a vicious circle is formed when the sea ice melts. Normally, the white ice reflects sunlight, which then bounces out into space, but when the sea-ice cover shrinks, the amount of sunlight reflected is also reduced. Instead, a larger proportion is absorbed by the surface of the ocean, which causes warming that contributes to the rise in air temperatures around the Arctic.
On the one hand, the rising temperatures make vegetation grow more vigorously and therefore more carbon dioxide is taken up, which is a positive effect. On the other hand, the same temperature rise means that more carbon dioxide and methane are released from the soil, which has a strong negative impact on the climate, according to Dr Frans-Jan Parmentier.
In addition to the changes on land, the present study shows that there are a number of uncertainties surrounding the effects of the melting ice on the amount of greenhouse gases exchanged by the ocean through natural processes. Many of these marine processes are poorly understood in this context.
"We know very little about how the shrinking sea ice cover disturbs the balance of greenhouse gases in the sea in the long term," says Dr Parmentier.
The article has been published in the journal Nature Climate Change.

Bisphenol A exposure in humans may be too low to cause problems by mimicking estrogen
Feb. 15, 2013 ? A controversial component of plastic bottles and canned food linings that have helped make the world's food supply safer has recently come under attack: bisphenol A. Widely known as BPA, it has the potential to mimic the sex hormone estrogen if blood and tissue levels are high enough. Now, an analysis of almost 150 BPA exposure studies shows that in the general population, people's exposure may be many times too low for BPA to effectively mimic estrogen in the human body.
The analysis, presented at the American Association for the Advancement of Science's annual meeting by toxicologist Justin Teeguarden of the Department of Energy's Pacific Northwest National Laboratory, Richland, Wash., shows that BPA in the blood of the general population is many times lower than blood levels that consistently cause toxicity in animals. The result suggests that animal studies might not reflect the human BPA experience appropriately.
"Looking at all the studies together reveals a remarkably consistent picture of human exposure to BPA with implications for how the risk of human exposure is interpreted," said Teeguarden. "At these exposure levels, exposure to BPA can't be compared to giving a baby the massive dose of estrogens found in a birth control pill, a comparison made by others."
In addition to evaluating the likelihood of BPA mimicking estrogen in humans, Teeguarden also analyzed another set of BPA studies that looked at the chemical's toxicity in animals and cells in the lab. These 130 studies are significant as a group because they refer to the exposures as "low dose," implying they are very relevant to human exposures.
According to his analysis, however, the "low doses" actually span an immense range of concentrations, a billion-fold. In addition, only a small fraction of the exposures in these self-described "low dose" studies are in the range of human exposures, from 0.8 percent to 7 percent depending on the study.
"The term low-dose cannot be understood to mean either relevant to human exposures or in the range of human exposures. However, this is in fact what it has come to mean to the public, as well as many in the media," said Teeguarden.
Analysis of 150 Exposure Studies
The first analysis covered 30,000 individuals, including women and infants, in 19 countries. Human blood concentrations were calculated multiple ways using many kinds of exposure data.
Teeguarden looked to see if BPA concentrations were sufficiently high to be a significant source of estrogen-like activity in the blood. Researchers have long known that BPA can bind to the same proteins that estrogen does -- called estrogen receptors -- when estrogen is doing its job in the body. However, in most cases, BPA does so much more weakly than estrogen. To trigger biological effects through receptors, BPA concentrations have to be high enough in the blood to overcome that weakness.
"Systematically testing the estrogenicity, or the bioactivity of BPA at the part per trillion concentrations we expect in human blood would seem the most scientific way to substantiate or refute this conclusion," said Teeguarden.
Teeguarden analyzed the data in these studies using multiple independent approaches applied systematically to the data from thousands of individuals. The results showed that human blood levels of BPA are expected to be too far below levels required for significant binding to four of the five key estrogen receptors to cause biological effects.
Teeguarden's analysis also confirmed the findings of many academic and government scientists that biologically active BPA is at such low concentrations in the blood that it is beneath toxicologists' current ability to detect it, raising questions about the role of sample contamination in studies reporting high levels of BPA.
Analysis of 130 Toxicity Studies
In this analysis, Teeguarden compiled all the BPA studies that included the term "low dose" as it referred to human exposure by using such terms as "low-concentration," "environmentally relevant," or "human exposure." From the 130 studies found, he and PNNL biologist Sesha Hanson-Drury compiled all the doses that were actually used in the studies.
The results showed that a small fraction of the "low doses" used in these studies are within the range of human exposures, with the vast majority being at least 10 to thousands of times higher than what humans are exposed to daily. In addition, the range of concentrations spans from upwards of 10 grams per kilogram of weight per day down to 100 picograms per kilogram of weight per day (a picogram is one millionth of a gram).
"Unfortunately, the low dose moniker has been used by some to promote the importance of selected toxicity studies, for example, in arguments to ban BPA," said Teeguarden. "For BPA and all chemicals, we need more accurate language to present these findings so the public and scientists in other disciplines can understand how human exposures compare to exposures in laboratory studies reporting toxicity."

On Friday, out of the blue, the Colts announced that they have parted ways with veteran defensive end/linebacker Dwight Freeney.? Technically, they haven?t.? Not yet.
More specifically, the Colts have decided that Freeney won?t be re-signed, which means that he?ll become a free agent on March 12.
Owner Jim Irsay gushed about Freeney in a release confirming the move, saying that ?[f]ew people have meant as much to the success of the Indianapolis Colts as Dwight Freeney,? that ?[h]e has been a dominant player,? and that he ?was an artist, a joy to watch, and the dedication he put toward his craft was a rare quality.?
Irsay also said that Freeney will be inducted into the team?s Ring of Honor.
But if the Colts and Irsay truly want to honor Freeney, they?ll do the honorable thing and cut him.? This would allow Freeney to have a three-week head start on free agency, searching for a team without having to box out the other various free agents who will all be chasing the same cap dollars.
If the Colts are, as they have declared, moving on, there?s no reason to squat on Freeney?s rights any longer.? So if they mean all those nice things they?re saying about Freeney, they?ll do something that will cost them nothing ? while also giving real meaning to the phrase ?part ways.?

"It's about diversity because we are such a wide range of people," Mack says. Given the HIV-related health struggles of Dot's owner peter Taylor he says the float is also about the triumphs of the human spirit.
The parade kicks off at the south end of Ponsonby Road at 4pm but organisers are advising spectators to arrive well before this time to find parking and good viewing positions.

What does a company do when its owners don?t believe in contraception yet healthcare regulators are mandating that contraceptives and birth control procedures be offered to all employees under their health care plan?
They take the matter to court?that?s what!
And that?s exactly what Indiana-based company, Grote Industries, did to win temporary court relief from providing staff with coverage for contraceptives, birth-control methods, and sterilization procedures, as mandated under the 2010 U.S. healthcare overhaul.
The vehicle safety systems manufacturing company, based in Madison, Indiana, is privately-owned by the Grote family. The Grote family claims in their court injunction that they are Catholic, and according to their belief system, therefore don?t morally agree with the contraception (i.e., such as oral birth control pills) and sterilization procedures (i.e., Vasectomies) listed under their group insurance plan.

A cure for miscommunication: Spanish for health care workers
William C. Harvey says he's "out of the cave" and ready to resume his mission: to teach Spanish to just about everyone in Orange County.
Harvey, 59, wrote the indispensable "Spanish for Gringos," published in 1999, and since then has authored more than a dozen other books on language instruction. Many of them are designed to help native English speakers in a specific profession get by with just enough Spanish to serve customers. Or in some cases, save a life.
William C. Harvey will teach the class "Spanish for Healthcare Professionals" through UCI Extension for five weeks, from Feb. 21 through March 21.

On Feb. 21, he will begin teaching a five-week course called "Spanish for Healthcare Professionals" at UC Irvine.
The class is offered through the school's continuing-education arm, UCI Extension, and will be held Thursdays 6:30-9:30 p.m. through March 21 at Social Science Plaza A, Room 1165.
The cost is $385 and there are seats left among the 35 spots.
Who should take this class? Doctors, nurses, desk clerks, support staff ? anyone who works at a hospital, HMO or clinic who would find a basic knowledge of Spanish useful to give or receive information.
"The program allows people who've never spoken Spanish before to stand before a Spanish speaker, understand the general message that's coming at them and respond intelligently," Harvey said.
The textbook used for the class will be Harvey's own book, "Spanish for Health Care Professionals," published in 2008.
His other books include "Spanish for Law Enforcement Personnel," "Spanish for Attorneys and Paralegals" and "Spanish for the Construction Trade," as well as a few books for native Spanish speakers on how to learn English.
Harvey doesn't get bogged down with perfect conjugation of verbs. The goal is to be able to connect with someone "warmly, culturally."
After his surge of prolific writing and speaking, Harvey said, "I still have that passion. I'm out of the cave now, letting the industries know I'm out there again."

Obama calls NKorea nuke test 'highly provocative'
WASHINGTON (AP) ? President Barack Obama on Tuesday called North Korea's latest nuclear test a "highly provocative act" that threatens U.S. security and international peace.
"The danger posed by North Korea's threatening activities warrants further swift and credible action by the international community," Obama said in a statement issued early Tuesday. "The United States will also continue to take steps necessary to defend ourselves and our allies."
In a statement, the director of U.S. National Intelligence estimated the explosive yield at "approximately several kilotons" and said the government was analyzing further details. The statement said the explosion occurred in the vicinity of P'unggye, which is a mountainous region that has been the site of previous nuclear tests.
North Korea said it successfully detonated a miniaturized nuclear device at a northeastern test site Tuesday. South Korean, U.S. and Japanese seismic monitoring agencies said they detected an earthquake in North Korea with a magnitude between 4.9 and 5.2.
North Korea's official Korean Central News Agency said the test was conducted safely but with "great explosive power." It said the test is aimed at coping with "ferocious" U.S. hostility that undermines the North's peaceful, sovereign right to launch satellites. Last month, North Korea's National Defense Commission said the United States was its prime target for a nuclear test and long-range rocket launches.
"These provocations do not make North Korea more secure," Obama said. "Far from achieving its stated goal of becoming a strong and prosperous nation, North Korea has instead increasingly isolated and impoverished its people through its ill-advised pursuit of weapons of mass destruction and their means of delivery."
The chairman of the House Foreign Affairs Committee, California Republican Ed Royce, released a statement Tuesday calling on the Obama administration to "replace its failed North Korea policy" and issue "stringent sanctions" against North Korean leader Kim Jong Un's regime. "Otherwise, the grave North Korean threat to the region and the United States will only grow," Royce said.
The U.N. Security Council will hold an emergency meeting Tuesday morning on North Korea's nuclear test. South Korea's U.N. Mission informed reporters early Tuesday that the closed-door meeting will begin at 9 a.m. EST.
The White House said that Obama, who is scheduled to deliver his State of the Union address Tuesday evening, had already intended to mention North Korea in the speech, describing the North's action as part of a pattern of provocative behavior that has undercut its government's legitimacy.
"The president will say that the only way North Korea will rejoin the world community is if they stop these threats and live up to their international obligations," said Tommy Vietor, a spokesman for the National Security Council.
Senior White House adviser Valerie Jarrett said she expects the international community "will have a very firm response."
She said in an appearance on NBC's "Today" show that North Korea's move presents a threat to the region and to the United States. "It actually is not in the best interests of North Korea," she said.
On Monday, U.S. Secretary of State John Kerry discussed North Korea's "continued provocative rhetoric" in a phone call with China's Foreign Minister Yang Jiechi. That followed earlier conversations with Kerry's counterparts from Japan and South Korea, key U.S. allies in the region.
State Department spokeswoman Victoria Nuland said the three conversations "were remarkably similar" on the importance of implementing the commitments of a January U.N. resolution that toughened sanctions against Pyongyang and warned of "significant action" if it conducted a nuclear test. That resolution was supported by China, North Korea's only major ally.
On Tuesday, China expressed firm opposition to the test but called for a calm response by all sides.
